The working principle of the low-temperature freeze dryer is to freeze the dried items below the three-phase point temperature. The main components are divided into drying oven, condenser, refrigeration unit, vacuum pump, heating/cooling device, etc. After pretreatment, the materials are first frozen, then vacuum dried, sublimated and dehydrated, and then packaged in the post-treatment workshop. The heating system of the vacuum freeze dryer provides sublimation latent heat for materials, and the refrigeration system provides the required cooling capacity for the water catcher and drying bin. The quality requirements for freeze-dried products are unchanged biological activity, uniform appearance color, full shape, firm structure, fast dissolution rate, and low residual water.

The freeze-dried solid material presents a porous structure due to the sublimation of small ice crystals and maintains the original frozen volume. It is very easy to dissolve and recover after adding water. The temperature of the product is kept at a low temperature (generally lower than - 250 ℃) during the sublimation process. Therefore, the low-temperature freeze-drying machine is suitable for those heat-resistant substances, such as enzymes, hormones, nucleic acids The drying of blood and immune products is particularly suitable. The result of drying can discharge more than 95 ~ 99% water, which is conducive to the long-term preservation of products. The drying process of products is carried out under vacuum conditions, so it is not easy to oxidize.
The glass bottle will not break under uniform heating or cooling at a certain temperature, but it will break if different temperatures are applied at different parts to form a certain temperature difference. Therefore, to reduce the temperature difference of each part of the glass bottle, the freeze-drying curve reflects the reduction of the temperature line difference between the partition temperature curve and the sample temperature curve. The temperature difference between the sample temperature and the diaphragm temperature should be controlled to be less than 20 ℃, which not only shortens the freeze-drying cycle but also solves the phenomena of bottle breaking and bottom falling. Of course, broken bottles are also related to the quality of bottles and the performance of lyophilizers.

When freezing, free water forms ice, which occupies a large volume, and the gap formed after sublimation is also large. The useful components are dispersed with water vapor during sublimation to form Plush substances. Once air absorbs water vapor and disappears, an appropriate amount of filler should be added to solve it. When the product comes out of the box, there are bubble pit, dry shrinkage, collapse, cavity, and other defects. The reason is that the freezing temperature is too high or the time is too short, and the product is completely frozen, Or the temperature is too high and the pressure is too high in the drying stage. Caused by melting some products. The low-temperature freeze dryer vacuums and sublimates before it is frozen firmly, and the external pressure decreases rapidly, causing bubbling; When the melted product produces steam concentration, it will shrink; Some dry products are heated violently and melted, which will melt the dried objects into cavities and collapse. Therefore, it must be frozen, and the sublimation cannot exceed the eutectic point temperature and disintegration temperature of the product. During heat release, the temperature of the product itself drops sharply to reach the eutectic point temperature. The shelf layer in contact with the bottle bottom does not have the above evaporation and freezing characteristics, and the glass bottle can not withstand such a large temperature in a short time.
