Example of process flow exploration of pilot freeze dryer:

Take the freeze-drying process of myocardial peptide for injection as an example to explain the exploration of freeze-drying process. Various factors that may affect the lyophilization process of myocardial peptide for injection were experimentally studied with a pilot lyophilizer (20 pieces per batch). A preliminary optimized process route was obtained by studying the heating rate and form in solution preparation, pre freezing and freeze drying. The original freeze-drying cycle was shortened from 78 hours to 45 hours. At the same time, the quality deviation in the batch was reduced from 2% to 1.5%. Through many exploratory experiments, it is found that the four factors that have the greatest impact on the freeze-drying process of myocardial peptide for injection: filling amount, feeding mode, slow period of one-time sublimation and high-speed period of one-time sublimation. 

Next, several exploratory experiments were carried out on the four factors of vacuum degree, air content, slow period of one-time sublimation and high-speed period of one-time sublimation to investigate their synergistic effects on the freeze-drying process of myocardial peptide for injection, and a further optimized freeze-drying route was obtained. After that, the process was applied to the pilot experiment (300 pieces per batch), and the pilot level was further adjusted and optimized. Finally, the process was applied to industrial scale-up (10000 pieces per batch), and the problems of long production cycle and large quality deviation in the batch were successfully solved.
