The freeze-drying extraction method of active milk protein in yak milk by freeze dryer includes degreasing, high temperature, and short-time sterilization, acid flocculation, ultrafiltration, vacuum freeze-drying, and other steps. The invention directly uses the characteristics of the high protein content of yak milk and the acid coagulation property of protein to extract the active milk protein. By extracting the active milk protein and using the principle of different flocculation of different acidic substances on different proteins, the active milk protein with high lactoferrin content is extracted and the activity of milk protein is retained, And the extraction rate of active milk protein in yak milk was improved to the greatest extent.

It includes the following steps:

1) Degreasing of yak milk: select pollution-free fresh yak milk with a protein content of 5.5-7%; Preheat to 38 ℃ and separate by 7000r / min centrifuge for 15 minutes to obtain skimmed milk and fat;

2) High temperature and short time sterilization: sterilize skimmed milk at high temperature and a short time at 80 ℃ for 15s;

3) Acid flocculation: add 6% acetic acid to sterilized skimmed milk, adjust pH to about 5, then add 4% citric acid to adjust pH to about 4.5, and control the temperature within the range of 25-35 ℃;

4) Ultrafiltration and vacuum freeze-drying: the concentrated protein solution is obtained by ultrafiltration. The ultrafiltration condition is that the flocculation solution is diluted twice at 25 ℃ and ultrafiltrated through the ultrafiltration membrane with a molecular weight of 10000. The finished product of active milk protein is vacuum freeze-dried at a vacuum chamber pressure of 0.1kpa and temperature of 10 ℃.
