Usually, the immediate test effect of lyophilized reagent is very good, but after a short period of time, we will find that the activity attenuation is very serious or the lyophilized reagent is inseparable from the cold chain. 1. Excipient formula: for lyophilized reagents, the shape and firmness of structure is one of the basic elements of the stability of lyophilized products.

2. Internal stress

The "stress" in the reagent has not been eliminated. Stress is a vivid metaphor in Materials Science: when you take a piece of paper, group it up and put it down, it will stretch and change slowly, and finally stabilize, and the shape will change.

During freeze-drying, there are also many stresses in the morphological changes of liquid reagents, especially for the products directly freeze-dried with plastic kits. The poor heat transfer efficiency leads to a large temperature difference, which will lead to a large internal stress. If these are not removed after freeze-drying, they will be slowly released later, resulting in instability. Usual practice: annealing is required when freezing and at the end of freeze-drying. During pre freezing, try to build internal stress formation, that is, the temperature difference before product freezing.

How to do annealing? The eutectic point and eutectic point were measured, and the annealing temperature was lower than the eutectic point.

Annealing can not only increase the stability of products, but also improve the sublimation efficiency of products. Of course, annealing also takes time, which is part of the work that freeze-drying process researchers must explore. Annealing at the speed of freezing and final analytical drying together to determine the withstand temperature of the product after freeze-drying. Do not misunderstand it as the temperature in liquid state. The withstand temperature of freeze-drying is higher than you think. This is not only a process of removing bound water, but also an annealing process.

3. Moisture content

How much water is left in the product? Is the moisture content of the products in the whole freeze-drying box qualified? How to judge the end of freeze-drying?

Some manufacturers use product temperature probes. In fact, there are many uncertain factors. The results will vary greatly with different probe positions. Pressure rise test is the best method. Close the main valve of the cold trap and judge by the speed of vacuum pressure rise. The uncertain factors of this method are the processing level of the lyophilizer and the aging of the sealing ring. The leakage rate of the lyophilizer itself needs to be maintained by the user. It is ideal to test the leakage rate before use.

4. Other factors

Finally, from production to packaging, set some conditions to ensure physical, chemical and biological factors, such as preventing moisture absorption and pollution before packaging, packaging sealing design and so on.
