Annealing can promote crystal growth, expand sublimation channels, reduce sublimation resistance, shorten freeze-drying time, and easily obtain an elegant appearance. There are two changes in the annealing process. When the temperature is higher than a certain temperature, the viscosity of the amorphous substance will decrease with the increase of temperature, so that flow and polymerization will occur; for the crystalline substance, the small ice crystal molecules will migrate to the large ice crystals.

During the freezing process of the solution in the lyophilizer, there is a phase separation process between the solute and the solvent. Most lyophilized medicines use water as the solution, so water is used as an example to illustrate the condensation process of the solution.

As the temperature drops to a certain temperature, water begins to crystallize, and the temperature at this time is the supercooling temperature of the product. Due to the exothermic heat of crystallization, the temperature of the product begins to rise and then decreases. As the water crystals increase, the concentration of the solution increases (in order to facilitate drying, generally freeze-dried product solutions are prepared as a dilute solution of 4%-15% solid matter).

In addition, depending on the nature of the product, there are two situations at this time:

1) The product whose solute can crystallize, as the temperature of the product drops, the dilute solution becomes a concentrated solution, and gradually becomes a saturated solution. When the temperature continues to decrease, due to the decrease in solubility, solute will be precipitated, and finally it will become a eutectic mixture of ice crystals and solute crystals. The size of the crystal is related to the freezing rate, and the temperature at this time is the eutectic temperature of the product. This process is also an exothermic process, and overcooling and plateaus also appear on the freezing curve.

2) For products where the solute does not crystallize, as the temperature of the product decreases, the dilute solution becomes a concentrated solution. When the temperature continues to decrease, the solution freezes in a glassy form. The temperature at this time is the freezing temperature of the product. A microscopic glass structure is formed in the gaps between ice crystals.
