The freeze-drying process is actually the physical state change process of the water in the product, and it is also the water transfer process, and the water transfer can also be called the mass transfer process. In the pre-freezing stage, the water changes from liquid to solid, which is an exothermic process, and the released heat is taken away by the freezer. In the sublimation stage, water is directly changed from solid state to vapor state, which is an endothermic process, and heat is provided by an electric heater. The sublimated water vapor flows from the freeze-drying box to the cold trap. This is the mass transfer process. The water is transferred from the inside of the product to the surface of the cold trap. This is the drying process. The water vapor condenses into frost on the surface of the cold trap. Water changes from a gaseous state to a solid state, which is an exothermic process, and the heat released is carried away by the freezer. After freeze-drying, the cold trap is defrosted from solid to liquid, which is an endothermic process, and heat is provided by defrosting water or steam.

The technological process of freeze-drying for modern pharmaceuticals is as follows: first, the freeze-drying box is cleaned, followed by fire bacteria. After the fire bacteria, leak rate detection should be done. Leak rate detection proves that the system has a good vacuum before the product can be put into the box. If the lyophilizer does not have a cleaning-in-place and sterilization-in-place system, it needs to be cleaned manually and sterilized by other suitable methods.
After the product is sub-packed into the box, pre-freeze the product, sublimation drying (first stage drying) and desorption drying (second stage drying), about 1 hour before the end of pouring and freezing, the cold trap should be cooled in advance to - The low temperature below 40℃, then start the vacuum pump, evacuate the cold trap and the freeze-drying box. When the vacuum of the freeze-drying box reaches 01 mbar, the sublimation starts, and the product is heated. Drying ends until the product reaches an acceptable residual moisture content.
After the drying of the product is completed, a vacuum or an air tamping is performed as required. If it is a vacuum plug, it should be carried out immediately after the drying. If it is a nitrogen filling plug, it needs to be pre-vented to make the ammonia gas charged to the set pressure, generally around 500~600 mmHg, and then pressurized. After the plug is finished, the air is released to the atmospheric pressure and out of the box. After the box is released, the aluminum cap is pressed, labelled, and packaged to become the product to be inspected. After passing all required inspections, the product becomes a finished product. The filter integrity in the inspection refers to the sterile filter of the air inlet of the freeze-drying box. If the integrity test of the air inlet filter fails, the batch of products is a waste product, so some freezers are equipped with two air inlets. Filters, used in tandem: The chance of two filter integrity tests failing at the same time is extremely small.
