Surgery is still the main solution for vesicovaginal fistula, but postoperative wound healing represents a challenge, and the recurrence rate remains high. There is a need to develop therapeutic methods to increase the success of therapy and women's quality of life. To explore whether human freeze-dried amnion is useful as a mesenchymal stem cell scaffold for repair of vesicovaginal fistula through assessment of the proliferative and remodelling phases by an amnion freeze drying machine.

Freeze-dried amnion was made by human amniotic membrane stem cells. Previously, dry amnion was prepared in 12 well plates obtained from the tissue bank, and was cut into 1*1 cm sizes. Each well was assigned Alpha medium–MEM. Stem cells from the amniotic membrane were prepared from one large plate (2*106 cells), taken using a Pasteur pipette, inserted into the tube, rinsed with phosphate buffered saline, given trypsin, then put into a 16-ml Falcon tube. A deposit of stem cells collected at the bottom of the tube. The sediment in stem cells that had been given alpha medium-MEM and resuspended was added to 1 ml of each well (2,000,000 stem cells/1 ml, according to the dose for rabbits), which already contained amniotic membrane scaffold. Twelve well plates containing stem cells that were seeded into this scaffold were subsequently placed in an incubator for 7 days.

Freeze-dried amnion was found to play a role in the wound-healing process in a vesicovaginal fistula repair model. It is hoped that this research will improve urogynaecological services. Experimental research focusing on the use of freeze-dried amnion in humans, and the other examination parameters related to the wound-healing phase in cases of vesicovaginal fistula is recommended.
